We have setup as below.
Two VM's one for OTRS (webserver) and second one for MYSQl
1. Two virtual machines are running on ESXi 4.0
2. RAM size of the host machine is 8GB and RAM usage is 2.6GB
3. Each virtual machine RAM size is 4GB
4. Network Speed is 100MB Full Duplex for all three machines
OTRS 3.0.9 ITSM 3.0.4 Installed. CMDB Around 5010 entries.
The system is very slow. It takes around 10 to 15 min for creation of ticket.

try running "suport assessment" and see where the bottleneck is

I dare say it's not problem of OTRS. It's a snap for creating a ticket in a second with OTRS.

- how is the system behaving while the ticket is created (which takes 10-15 mins)

- my 1st assumption would be: check your database regarding index, ...; i had a similar case where indexes where turned off for one table in mysql
- nevertheless: "Support Assessement" give you a lot of hints
